This is very strange.
I have been starting the scoot up over the Australian winter ON THE CENTER STAND and it revs out fine and all seemed ok. I went to take it for a ride and as soon as I put both wheels on the ground it started to backfire at the slightest of throttle openings and got worse with more throttle. Back on the center stand, no backfire. I pushed down on the back of the seat to elevate the front wheel to the same height as the rear, both wheels horizontal (while still on the center stand) and it revs out fine.
When I did take off on it, it coughed and spluttered and cut out. It can sit there idling fine for ages though
Any ideas what could be causing this?

did you check the vacuum line going to valve right hand side of carb for being anti back fire?

I pulled the reed block out and it is in order.
I was running a #90 main jet so I don't think it can be lean. I don't know how getting it off the stand would affect the richness/leanness. Plug is black.
I pulled the carby apart, cleaned everything, put an #80 mains in it while I was at it, still the same problem.
One other thing I did was put the rear brake on while it was on the center stand and revved it out. It revved ok, no backfire.

It may be an electrical problem,, I hate to say it but pull the plastics and check the routing of the wire harness,,, it could be getting pinched or a bad connector plug or one may not be seated correctly and its "pulling apart" slightly to give an intermittent connection

are you running it with out a exhaust pipe and muffler?

ok now the starter isn't working, no turn over of the starter motor just a click click (battery is charged tried another battery as well to make sure)

Cross the solenoid terminals with an old screwdriver if she turns over the solenoid is bad,,,,,if no turn over starters bad

Each is own but with out exhaust system for back pressure it will back fire and will never run 100 % right.

I crossed the terminals, sparks flew and then it blew the fuse.
P.S. Where is this no exhaust stuff coming from, the exhaust hasn't been off during this caper ....??
